Dissecting the regulatory mechanisms of alternative polyadenylation and nuclear retention of mRNAs by SR proteins

SR proteins are central RNA-binding proteins that accompany mRNAs throughout their journey from transcription in the nucleus to translation in the cytoplasm. They thereby hold the potential to link subsequent RNA processing steps for nuclear surveillance. However, their functions beyond splicing remain poorly studied. Here, we will elucidate the molecular mechanisms underlying the roles of SR proteins in alternative polyadenylation and regulated RNA export. We will study the physiological consequences of these regulatory functions in the context of neuronal differentiation.structure thus reflects its functional importance. We will use this structural conservation to identify previously unknown cis-regulatory elements and establish a high-throughput screen for the parallel analysis of thousands of conserved 3’UTR structures in different cell types and under different stress conditions. The importance of trans-acting factors known to recognize mRNA structures will be tested globally for all functional motifs, and the best candidates characterized in detail. More
